Cannabis, anxiety, and depression have a close, complicated relationship. While cannabis use can be driven by its anxiolytic effects, tetrahydrocannabinol (THC) itself can induce panic and anxiety particularly at high doses anartd in susceptible individuals. Similarly, cannabis use can reduce symptoms of depression and elevate mood, but can also predispose individuals to the development of depression. This is particularly true in adolescents and those who use cannabis heavily. Although we know a considerable amount about the effects of THC and other CB1 receptor agonists in preclinical models of anxiety and depression, we know very little about other phytocannabinoids or about combinations of phytocannabinoids. Phytocannabinoids also affect the body through alterations in key endocrine systems and thereby widely influence homeostatic processes. While considerable data are available regarding the effects of THC on the hypothalamic–pituitary–adrenal axis, very little is known about its effects on other hormonal systems and virtually nothing is known about other phytocannabinoids.
