This chapter discusses the physiologic link between external exposures and internal tissue concentrations for an exposed individual. It is possible to construct simplified descriptive toxicokinetic models formalizing a reasonable inference about key kinetic processes for many environmental chemicals. These can give insight on the ways materials move through the body and are processed. This is because the possible routes of entry, possibilities for metabolism, long-term storage and removal pathways are limited, and depend strongly on the biochemical and physiological characteristics of the substance. Simple toxicokinetic models allow us to visualize how variation in exposure affects the tissue concentrations over time. They also permit us to adjust our dose metrics to compensate for differences in body size, gender, age, and some genetic factors.
