The neuromuscular junction nAChR undergoes a developmental switch around the time of birth in which expression of the γ subunit is replaced by expression of the ε subunit, but otherwise the vertebrate muscle nAChR forms an essentially homogeneous receptor population of fixed subunit composition. There is now evidence for the assembly of neuronal nAChRs containing only one type of subunit (homomeric receptors) and also for those containing two, three, and four different subunit subtypes (heteromeric receptors). It appears, however, that all nAChRs contain a total of five coassembled subunits. This chapter presents a schematic representation of the pentameric arrangements of subunits in an assembled nAChR, together with an illustration of the putative membrane topology of individual nAChR subunits.
