This chapter examines some of the most important studies on the topic of structural plasticity in dendrites and spines, placing them into the context of information storage in the brain. The chapter starts with an assessment of the classical literature, which mostly relied on conventional light and electron microscopy. However, most emphasis in this chapter is on studies using state-of-the-art imaging approaches, which allow microscopical structural changes to be followed over time, including in awake animals performing complex behavioral tasks. The chapter ends with five particularly important open questions, providing an outlook into the future, and a discussion of where research on structural plasticity in relationship to synaptic plasticity and memory could stand in a few years’ time.
